หลังจากที่ถูกแมงมุมกัมมันตภาพรังสีกัดในระหว่างที่เขากำลังศึกษาการทดลองทางวิทยาศาสตร์ ในชั้นเรียนอยู่นั้น ปีเตอร์ ปาร์คเกอร์ ก็ได้พบกับสิ่งแปลกใหม่บางอย่างที่เข้ามาในร่างกายของเขา นั่นก็คือพลังพิเศษที่เขาได้รับจากแมงมุมตัวนั้นนั่นเอง
